Overview
I have keen interest in understanding the molecular pathogenesis of osteoarthritis (OA) and identifying and targeting potential therapeutic targets to stop/delay the joint inflammation, and cartilage destruction to restore joint function. We used several research tools employing cellular, molecular, biochemical, high throughput sequencing and system biology approach in the in vitro cell culture model and in vivo genetic animal models. My research discovered the role of an orphan GPCR as a novel regulator of inflammation and apoptosis in OA. Using CRISPR/Cas9 based genome editing and long-range interaction mapping by 4C-sequencing in stem cells (iPSCs), we have identified the role of regulatory sequences within intronic region of RUNX1 locus for skeletal cell differentiation and bone formation using Calvarial defect model in mice. My other research focus involved regenerative medicine approach for cartilage preservation, repair and regeneration of joint tissue using stem cells (MSCs) in various model of post-traumatic osteoarthritis (PTOA) in mice and rats.
