Overview

Dr. Sorscher has maintained a research focus on cystic fibrosis (CF) related science and clinical intervention for more than 25 years. He has contributed to studies of cellular and molecular biology describing aminoglycoside suppression of CFTR truncation alleles, clinical significance of mild CFTR mutations and the bioelectric phenotype, acquired loss of CFTR function in vivo, and mechanisms underlying activation of CFTR by potentiator and corrector compounds. In this manner, he has participated in recent advances with regard to CF molecular pathogenesis and experimental therapeutics. Studies of other orphan conditions are also pursued by Sorscher and colleagues using lessons learned from CF. For example, a major interest involves characterization of a mechanism for tumor sensitization using the E. coli PNP gene. In this approach, tumors are rendered hundreds or thousands of times more sensitive to conventional chemotherapy by expression of a prokaryotic enzyme that cleaves nontoxic nucleoside prodrugs to a very active form. The research involves analysis of the crystal structure of E. coli PNP, and structure-based drug design of new compounds that are effectively cleaved in vivo.
