Brief descriptions of all core services are listed below. If you would like additional information regarding these offerings, please contact us at mousecore@emory.edu.

Transgenic Mouse

Transgenic Mouse in FVB or C57BL/6

The core will release the transgene and purify it with the enzymes supplied by core user; we request that the PI provide 30-50 ug of plasmid DNA to the core, along with a plasmid map clearly indicating the relevant restriction enzyme sites.

Our guarantee: at least two transgenic founders per project.

Note 1: the core doesn’t guarantee transgene expression.

Note 2: The investigator should provide clear genotype result within 3.5 weeks after the pups are born, animal per diem fee will be billed to the PI account after 4 weeks of pups’ age.

BAC Transgenic Mouse

BAC constructs should be purified by the PI, preferably linerized and provided to the core at a minimum concentration of 10 ng/ul

Our guarantee: at least two transgenic founders per project.

Note 2: The investigator should provide clear genotype result within 3.5 weeks after the pups are born, animal per diem fee will be billed to the PI account after 4 weeks of pups’ age

ES Cell Gene Targeting

ES Cell Gene Targeting with HZ2.2 (129/SvEv/Tac)

The core will linerized, purify, and electroporate the construct into our stock ES cell line, HZ2.2 (129SvEv/Tac) and pick 200 drug resistant clones for expansion. Ethanol precipitated DNA (in 96 well formats) will be provided for analysis to identify recombinant clones. Up to 6 targeted clones will be expanded, DNA aliquot will be provided to PI lab for confirmation.

Our guarantee: we guarantee our best effort, even though we don’t guarantee homologous recombination event for every project due to locus specific nature of the procedure, however we have achieved high targeting frequencies in the past, please see our Success Rate.

Note 1: it’s the investigator’s responsibility to identify and confirm correctly targeted ES clones

Note 2: if no positive clones identified from the first 200 clones, the core will repeat gene targeting with newly prepared plasmid supplied by PI lab at no additional cost, however, additional fee will be charged for $1290 for each 96 clones after another 200 clones screened

ES Cell Gene Targeting With C57BL/6 ES Cell Lines

The core will linerized, purify, and electroporate the construct into commercial ES cell line (like KOMP’s JM8), and pick 200 drug resistant clones for expansion. Ethanol precipitated DNA (in 96 well formats) will be provided for analysis to identify recombinant clones. Up to 6 targeted clones will be expanded, and a second DNA aliquot provided for confirmation.

Our guarantee: we guarantee our best effort, even though we don’t guarantee homologous recombination event for every project due to locus specific nature of the procedure, however we have achieved high targeting frequencies in the past, please see our Success Rate.

Selection Marker Removal in Vitro

This service allows for the in vitro deletion of drug selection cassettes (or other target sequences) utilizing either cre or frt recombinase. Up to 12 drug sensitive clones are picked for each parental clone, and cell pellets will be provided to PI lab for analysis.

Our guarantee: a minimum of two properly deleted clones are guaranteed per parental clone.

Note 1: the PI lab should provide genotype result within 48 hours after cell pellets are provided, otherwise the core will charge $350 per clone after the first 6 clones are cryopreserved

Note 2: it’s investigator’s responsibility to identify and confirm correct clones (marker removed)

Knock-Out/Knock-In Mice Production

Correctly targeted and confirmed ES cell clones generated within the core are injected into blastocysts to generate chimeric mice. Up to two independent clones are injected for each project.  High percentage chimeric males are bred to wild-type females for potential germline F1 pups.

Our guarantee: we guarantee germline transmission for the construct targeted with our stock ES cell line, HZ2.2 (129SvEv/Tac), and at least 3 good chimeras for all other cell lines.

Note 1 : the PI lab is responsible for submitting ES cell clone ID before blastocyst injection and cofirm germline transmission

Note 2: per diem will be charged to PI account after 4 months passed blastocyst injection

Chimeric Mice Production (ES Cells Supplied From PI)

This service is aimed to generate chimeric mice from ES cell clones provided to the core from outside sources (other core facilities or consortiums like EUCOMM, KOMP...). The core will expand up to two clones per project, perform pathogen screening analysis (required), and inject the clones to appropriate recipient blastocysts.

Our guarantee: we guarantee our best effort, even though we don’t guarantee germline transmission, however we have achieved 100% germline transmission rate in the past 3.5 years. Please see our Success Rate.

Note 1: the PI lab is responsible to acquire ES cell karyotype information

Note 2: extra $550 for the third ES clone to make a total of 3 clones injected per project

Note 3: per diem will be charged to PI account after 4 months passed blastocyst injection

Cryopreservation Services

Embryo Cryopreservation with mice provided from PI lab

Valuable mouse lines are cryopreserved as 2-cell stage embryos. The core will produce embryos from up to 14 mating pairs of the desired line, and freeze all embryos that are collected. A single straw will be thawed per project and tested for viability. Fifteen embryos are frozen per straw, and can be stored by the core at no additional cost.

Our guarantee: we test thaw frozen embryos to live born pups.

Note 1: to start the project, we need at least 10 breeding pairs with young females and proven male breeders. PI lab is responsible to genotype the live born pups.

Note 2: if not enough embryos collected from 2 sessions, the PI will pay for a new fee

Embryo Cryopreservation via IVF

The core will produce 2-cell stage embryos from IVF (In Vitro Fertilization) with stud males provided from PI lab, and freeze all embryos that are collected from one IVF session. A single straw will be thawed per project and tested for viability. Fifteen embryos are frozen per straw, and can be stored by the core at no additional cost.

Our guarantee: we test thaw frozen embryos to live born pups.

Note 1: we need 2-3 stud males preferably those gave recent litters, 16 wt-female mice will be purchased from PI account per project. PI lab is responsible to genotype the live born pups.

Note 2: if not enough embryos collected from two sessions, the PI will pay for a new fee

Sperm Cryopreservation with IVF as a QC

Valuable mouse lines are cryopreserved as sperms. The core will freeze sperms in 12 straws per project, a single straw will be thawed per project and tested for viability by IVF, and straws can be stored by the core at no additional cost.

Our guarantee: we test thaw frozen sperm for IVF to live born pups.

Note 1: we need 3-4 stud males preferably those gave recent litters. 9 wt-female mice will be purchased from PI account per project for IVF. PI lab is responsible to genotype the live born pups

Rederivation from Frozen Embryos

We rederive frozen embryos to live pups came from external sources or the ones we froze.

Our guarantee: we guarantee live born pups from the embryos frozen by us.

Note: the embryos can’t be shipped until the health report is approved from Emory DAR, please contact Cherie Cosby Lawley before arrange the embryo shipment.

Through IVF, we rederive frozen sperms to live pups came from external sources or the ones we froze.
